Hi mates is there any propper way to configure a policy in Harmony Endpoint that block almost all the usb ports and left 1 port unblocked?

I don't believe so.
I haven't been able to find any settings that influence specific USB controllers/ports, just a general policy.
It would be difficult to manage across devices as well; not every device comes with the same USB port configuration.
And I'm not sure what you'd actually accomplish through this
I think your best bet would be to do something within the BIOS. Sometimes the BIOS will allow you to disable/enable certain ports and most manufactures provide tools that allows you to configure the BIOS remotely or using a script.
What @Swiftyyyy said is accurate. This is not possible using MEPP (Media Encryption and Port Protection) and also would be very difficult and nearly impossible due to trying to do this with different makes and models of machines.
Usually, hot glue is used to achieve this ! 🤣

You could take it as a joke, but this is a valid method. Only drawback is that you have no revert option 8)

If you have some trust in your users you could seal it only - stuff a paper bit in to save contacts and seal with hot glue. This is removable.
